Growth Hormone Releasing Peptide 6, GHRP6
GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide with the sequence His-D-Trp-Ala-Trp-D-Phe-Lys-NH2. It was one of the first synthetic GH secretagogues developed and remains widely used in research.
GHRP-6 stimulates growth hormone release by binding to the ghrelin receptor (GHSR) in the pituitary gland. Unlike hexarelin, GHRP-6 also significantly stimulates appetite through ghrelin-like effects, which may be desirable or undesirable depending on research goals.
This peptide produces a robust but not excessive GH pulse, making it useful for studying the effects of physiological GH elevation.
GHRP-6 acts through several mechanisms:
- Ghrelin Receptor Agonism: Binds to GHSR-1a, the ghrelin receptor, on pituitary somatotrophs.
- Somatostatin Suppression: May suppress somatostatin release, removing GH inhibition.
- Hypothalamic Action: Stimulates GHRH release from hypothalamus.
- Appetite Stimulation: Activates hunger pathways through ghrelin-like action.
- Gastric Effects: Increases gastric motility and acid secretion.
GHRP-6's effects on GH release have been extensively studied:
- Pulsatile Release: Produces physiological GH pulses rather than sustained elevation.
- Synergy with GHRH: Combined with GHRH, produces synergistic GH release.
- Age Effects: Maintains efficacy in aging subjects with blunted natural GH release.
- Consistent Response: Shows less receptor desensitization than hexarelin.
GHRP-6 remains valuable for GH physiology research.

GHRP-6's appetite effects have research applications:
- Ghrelin Mimetic: Mimics the appetite-stimulating effects of ghrelin.
- Caloric Intake: Studies show increased food consumption after administration.
- Gastric Function: Increases gastric motility and secretion.
- Cachexia Research: May have applications in wasting conditions research.
The appetite effects may be beneficial or limiting depending on research context.
